What is MOTS-c?

MOTS-c is a 16-amino-acid peptide encoded within mitochondrial DNA. Research has focused on metabolic homeostasis, skeletal muscle, cellular stress and more recently innate immune signalling.

Proposed mechanism

MOTS-c can move from mitochondria toward the nucleus during metabolic stress and influence nuclear gene expression. Experimental work links it with AMPK-related metabolism, glucose handling, adaptive stress responses and immune signalling.

What has been published?

Most intervention evidence remains preclinical. Human studies have measured endogenous circulating MOTS-c, including responses to exercise and associations with metabolic phenotypes. Newer research also describes host-defense and immunomodulatory functions.

Current human evidence

Human evidence mainly shows that MOTS-c exists endogenously and changes with physiology. This is not the same as evidence that administered MOTS-c improves performance, obesity, diabetes or longevity.

Regulatory status

MOTS-c remains investigational and is not an FDA-approved drug treatment.
